Am i not right in assuming anyone else would try the mainboard for the described fault???? 🤔 If i had bought a part through ebay and blagged it didnt work i would just admit it was me but it wasnt. Im pretty sure that there will be lots more to be said on this fault in the ue40ku6000 & ue43ku6000 as there rubbish just like the ue40ku6400

If I were you I will put all the original boards back in place, then take the DCV reading on the connector of the power supply board that goes to the main board when the TV is OFF and when the power switch is activated. So you know what signals and Voltages are missing or not correct.
At least the old board sends the BL-ON signal to turn on the backlighs but you sub boards do not even do that.
Once you get the reading of the old board, then you will put in the sub board in place and find out what DCV you have on the connector so you can at least compare the readings to the original board.
We also need good clear clean straight shot pictures of the whole back side of the TV.
What is the P/N of the power supply board? BN44-??????.
Make sure to report the DCV reading in the format that has pin name and follow by the DCV readings.
Pictures of the original main board and the sub boards? Especially the PCBA numbers as printed on the bard code stickers.
BTW, the board can be same blank PCB but it can be stuffed with different parts to make many version of the PCBAs. That is why always go by the numbers on the barcode stickers.

Nope, not without troubleshooting the original main board first. There are a few measurements to take, maybe even the oscilloscope to drag out. Replacing boards to get something back working, my 10 year old son can do that by himself, while the 8 year old is passing him the screwdriver.
Certainly I wouldn't open multiple working other TV's to try boards.
I wouldn't want a new TV that had been previously opened by a not certified tech, monkeyed around with and then sold. You wouldn't feel good either, if you buy a brand new car and someone has taken the engine out and put back in, only to test it in another car to see if it works. I am not sure how you get these many new TV's with these problems, but that isn't my beer anyhow.
