Did I need connect power to the PCB/TV or all needed powers for EN25Q40B are coming from CH341A programmer?
I know how to mod CH341A to work fully on 3.3V, so 3.3V must be not a problem.

There is no need to supply power, everything is powered through the programmer.

OK, I ordered CH341A and will first test writing EN25Q40B without soldering it out.
But on TV LED is blinking 1 long, 5 times short in cycle and as I understand no LED blink codes are available for that TV model and error can be also somewhere else than IC401 memory content fault.

I also checked voltages on board. 3,3V is present, 13V is present, 1,8V CPU power is present, but 5V USB power is missing and also back LED 43V is missing. Can faulty EN25Q40B content cause missing USB and back LED voltages?

The processor power supply cannot be 1.8v; it should generally be 0.9-1.0v. Due to damage to the standby firmware, the TV will not start and there will be no main power supply for operating mode...

It can by that 1.8 V is not CPU power, it was closest to CPU marked voltage pin on PCB without removing CPU heat sink.
So is it normal that USB 5V and back light LED 43V are missing when TV is not in operating mode and most probably problem is IC401 information damage and can be fixed with writing new firmware to it?

I find one more thing what can indicate what problem I have with bn91-21809k board. When after power on indication led is blinking 1 long, 5 short blinks, seems like actually TV is not in standby mode as main CPU heat sink temperature rises within 20 minutes to 38 C degrees. I understand in standby mode main CPU must not have power, or I am wrong?

Looks like you have other issues than the firmware corruption as if you read the first post you will see that the standby led flickers when the remote control is pressed. If that is not the issue then you have incorrectly diagnosed your issue with your TV. As you have ordered the ch341A programmer than you can always try to see if it resolves your issue. I would suggest you read the flash ic first to see if the data is corrupted. Good luck
