The voltages look normal.

i have a speaker connection on main board that works fine if i plug in headphone speakers
ive circled this in red

ive noticed i also have a strange ticking sound by the right speaker ?

Try isolating left and right speakers individually by cutting one wire on each. Then try with just one in.

I am not sure if the audio output at the headphone jack has its own amplifiers that are not the same Class D power amplifiers for driving the speakers.
I think power amplifier is in that section that has 4 inductors with 4R7 printing on the top.
Need to find the diagram for this main board next.

Probably a bad audio amplifier IC. I doubt a schematic for E-series (2012) Samsung will be available.
